A TS-style pedal is critical here—the Champion cannot produce Marshall-style breakup on its own, and the Tube Screamer's mid-hump directly addresses the Fender's mid-scoop while adding the compression Slash's tone relies on.

PRO TIPRoll your Strat's tone knob to 6 and use the neck pickup to approximate the darker, thicker 'woman tone' vocal quality of the Les Paul's Alnico II humbucker—this is how Slash gets that singing sustain on the intro riff. Dig in with your pick near the bridge for attack, then move toward the neck for the smoother solo sections.
