Why these settings

Your HT5R's ISF should sit around 2 o'clock (toward British) to add some EL84 chime that complements the Fender-style mids SRV got from his Vibroverbs. The Blackstar's overdrive channel is gainier than a cranked Vibroverb, so gain 5 (not higher) paired with the TS9 boost gets you into that singing sustain territory without mud. Your Player Strat's pickups are hotter than SRV's '59 units — treble 6 compensates. The HT5R MkII has no presence control, so that's null.

PRO TIPSRV held his pick sideways (nearly perpendicular to the strings) and attacked hard with his thumb partially muting — this gave his solos that aggressive 'bark' on the attack followed by smooth sustain.
