Why these settings

Plugging your single-coil Strat into the High Sensitivity input requires pushing preamp gain to 8 and keeping bass at 7 to compensate for single-coil thinness. Treble is dialed back to 5 and presence to 4 to tame the inherent 6kHz harshness of ceramic Strat pickups through a JCM800. If you acquire a TS-style overdrive pedal later, set its Level high and Drive low to tighten the bottom end and push the JCM800 preamp into full saturation.
